چکیده

The transforming growth factor βs (TGF-β) belong a family of structurally related, dimeric, disulfide-linked polypeptides that includes five TGF-β isoforms, activins, inhibins, Müllerian inhibiting substance, bone morphogenic proteins, and products of the Xenopus Vg1 and Drosophila decapentaplegic (dpp) genes (reviewed in Rizzino, 1988; Massagué, 1990; Roberts and Sporn, 1990). In mammals, three isoforms of TGF-β (TGF-β1,-β2, and β3) have been identified. TGF-βs are multifunctional cytokines that influence numerous cellular processes. They can regulate cell proliferation and differentiation positively or negatively depending on cell type and have been implicated in such diverse physiological events as angiogenesis, immune function, steroidogenesis, and tissue remodelling and repair. Since all of these events take place in the uterine endometrium during the oestrous or menstrual cycle and during the establishment of pregnancy, TGF-βs are likely candidates to regulate some of these reproductive functions. Biological effects of TGF-β are mediated through interactions with cell surface receptors, designated type I, II and III (reviewed in Massagué and Weis-Garcia, 1996). Type I and II receptors are transmembrane proteins containing cytoplasmic serine/threonine kinase domains. TGF-β binds the type II but not type I receptor. The type I receptor recognizes the ligand bound type II receptor with which it forms a heteromeric complex. The type I receptor then undergoes phosphorylation, which is probably catalysed by the type II kinase. Signalling downstream of the type I receptor involves the phosphorylation of one or more cytoplasmic proteins referred to as Smads, complex formation of Smad-2, -3 and -4, and their translocation to the nucleus and association with transcription factors. The type III receptor is a proteoglycan with no cytoplasmic kinase domain and may be involved in presentation of TGF-β to the type II receptor. P-217: Expression Analysis of The Histone Variant H2A.Z in Endometrium Tissue during The Menstrual Cycle

Background: The human endometrium undergoes cyclical changes including proliferation, differentiation, tissue breakdown and shedding (menstruation) throughout a woman’s reproductive life. The postovulatory rise in ovarian progestrone induces profound remodeling in chromatin structure of cells, and consequently differentiation of estradiol-primed endometrium. This change is crucial for embryo im...

P-76: Important Role of Macrophage Colony-Stimulating Factor during Menstrual Cycle and Early Pregnancy Phase in Oovarian Stimulated IVF-Ppatients

Background: M-CSF is a cytokine mediating the growth, proliferation and differentiation of various cell types including macrophages, trophoblast cells, and osteoclasts. It plays an important role in immunity reproduction, follicle development and ovulation. In the present study, we describe the important role of the changes in serum M-CSF levels during the menstrual cycle, in the process of fol...

O-46: The Expression of Leptin and Its Receptorduring Different Physiological Stages in theBovine Ovary

Background: Leptin, the hormonal product of the obese (ob) gene, circulates in the blood at levels paralleling those of fat reserves and regulates satiety. In cattle, leptin has also been implicated in the control of ovarian function, but its local production in the ovary and role in the control of ovarian function in autocrine/paracrine manner is unknown. The aims of this study were to documen...

P-80: Histological Assessment and Oestrous Cycle Restoration after Vit-rified-Warmed Rat Ovarian Tissue Autotransplantation, Using Hyaluronic AcidHydrogel Scaffold

Background: Follicular pool is reduced after transplantation of vitrified ovary. Therefore, methods should be improved to accelerate ovarian function revival and maintain follicular reserve. In this study hyaluronic acid hydrogel as a scaffold was used to improve restoration of ovarian oestrous cycle. Materials and Methods: 79 mature (~8 weeks) rats having normal estrous cycles were divided int...

Differential regulation of interleukins IL-13 and IL-15 by ovarian steroids, TNF- and TGF- in human endometrial epithelial and stromal cells

Based on the endometrial spatial and temporal expression of interleukins (ILs) IL-13 and IL-15 during the normal menstrual cycle, we hypothesized that ovarian steroids and non-steroidal factors regulate their expression in a cell-specific manner. To test this hypothesis and determine IL-13/IL-15 actions, we used endometrial epithelial (EEC) and stromal (ESC) cells isolated and cultured under de...
